Common gut bacteria can fuel the growth of prostate cancers and allow them to evade the effects of treatment, a new study finds. Scientists revealed how gut bacteria contribute to the progression of advanced prostate cancers and their resistance to hormone therapy -- by providing an alternative source of growth-promoting androgens, or male hormones. Hormone therapy is the standard of care for advanced prostate cancer and works by lowering levels of androgens. But researchers found that low androgen levels in patients can drive the expansion of gut bacteria, which can become hormone factories to sustain prostate cancer growth. Bacterial 'fingerprints' identified by scientists may help pick out patients at high risk of developing resistance to treatment who could benefit from strategies to manipulate their 'microbiome'. Introduction Radical cystectomy (RC) is the standard treatment for certain stages of bladder cancer. For the high-risk group of patients with non-muscle invasive bladder cancer (NMIBC), both the European Association of Urology (EAU) and American Urological Association (AUA) guidelines recommend treatment with bacille Calmette–Guérin (BCG). 1,2 However, even in these patients RC is carried out. Yafi et al, for example, reported on 2287 RCs performed in Canada between 1998 and 2008, and the distribution of clinical staging was NMIBC in 26.3% (cTis in 4.8%, cTa in 2.9%, and cT1 in 18.6%). 3 The reasons for performing an RC include unresectable non-invasive tumors and BCG unresponsiveness. Indeed, the latter situation is one of the reasons for recommending RC. NEW YORK – Researchers from the Ontario Institute for Cancer Research have developed an algorithm that can better classify cells from the tumor microenvironment and provide tissue of origin based on single-cell transcriptomics data. In a talk at the annual meeting of the American Society of Human Genetics, held virtually last week, Ido Nofech-Mozes, a graduate student at the University of Toronto, described how his team developed a tool for the automated annotation of cancerous, immune, and stromal cells in single-cell RNA-seq cancer research. Manual annotation of these cells is difficult because a "large degree of interpatient heterogeneity in cancer cells makes it hard to find consistent markers that can be used across cancer samples," he said. "Cancer cells tend to cluster by patient and not by cell type, even," he said.
